The Sahlgrenska Academy, Department of Biomedical and Clinical Sciences, Division of Medical Chemistry and Cell Biology, Mitochondrial Genome Maintenance and Expression Group is recruiting a researcher for a temporary position. The group currently consists of approximately 15 researchers, including 4 PhD students and 2 principal investigators.   Duties  In this project, the candidate will investigate the biochemical and cell biological mechanisms that regulate mitochondrial DNA replication. The focus is on understanding how the involved proteins interact and how disease-causing mutations affect replication efficiency and genome stability. The work includes a combination of in vitro biochemical assays and cell-based methods. The applicant should have experience conducting research at an internationally competitive level in mitochondrial biology. Research experience will be assessed based on scientific publications. This postdoctoral position is part of a research project funded by a Wallenberg Scholar grant. The postdoctoral researcher will work in an interdisciplinary team with expertise in molecular biology, biochemistry and structural biology. The project aims to understand the molecular mechanisms that govern mitochondrial DNA replication and gene expression, and how defects in these processes contribute to human disease.   Subject area  Structural studies of mitochondrial DNA replication   Subject area description  Mitochondrial DNA replication is essential for maintaining mtDNA integrity and stability and thereby for normal mitochondrial function and cellular energy production. Defects in the replication process can lead to mtDNA depletion or deletions and cause mitochondrial disease. Our research aims to understand the molecular mechanisms that govern replication of human mtDNA. We combine structural biology, in vitro biochemistry and cell biology to investigate how the mitochondrial DNA replication machinery is organized and regulated. A particular focus is on structural and functional studies of the protein complexes involved in mtDNA replication and on understanding how disease-associated mutations affect these processes.   Job assignments In this project, In this project, the postdoctoral researcher will carry out structural and functional studies of protein and protein-DNA complexes involved in mitochondrial DNA replication. A central aim will be to determine structures of replication complexes using single-particle cryo-electron microscopy (cryo-EM) and use these structures to understand how the proteins interact and regulate mtDNA replication at the molecular level. The work will include preparation and biochemical characterization of protein complexes, cryo-EM sample preparation and grid optimization, as well as processing and structural analysis of cryo-EM data. Structural findings will be combined with in vitro biochemical analyses to test mechanistic models and investigate how disease-associated mutations affect the function of the replication machinery. The postdoctoral researcher is expected to drive the project independently while working closely with other members of the research group and our collaborators.   Eligibility  The eligibility criteria for employing teaching staff are set out in Chapter 4 of the Higher Education Ordinance and in the Appointment Procedure for Teaching Posts at the University of Gothenburg. To be eligible for appointment as a postdoc, the applicant is required to have a doctoral degree, a doctoral degree in art or a foreign degree that is deemed to be equivalent to a doctoral degree. Special grounds relate to leave of absence due to illness, parental leave, commissions of trust within union organisations, service within the defence services or other similar circumstances, as well as clinical service or service/assignment relevant to the subject area.    Assessment criteria  We are seeking a highly motivated researcher with a doctoral degree in structural biology, biochemistry, molecular biology or a related field. Documented experience in structural studies of proteins or macromolecular complexes using cryo-electron microscopy is required. Experience in protein purification and protein biochemistry is also required. Experience in single-particle cryo-EM, including sample preparation, grid optimization, data processing and structural analysis, is particularly meritorious. Experience in structural studies of protein-DNA complexes is a strong advantage. Knowledge of mitochondrial systems and experience in cloning, mutagenesis, protein expression, cell culture and in vitro biochemical analyses are considered merits. Great emphasis will be placed on scientific skills and the ability to independently plan, carry out and analyse research. The work involves close collaboration both within and outside the research group, making good teamwork and communication skills important. The candidate is expected to have strong scientific curiosity and the ability to work both independently and as part of an interdisciplinary team. Institutionen för molekylärbiologi söker en postdoktor för en tidsbegränsad anställning på två år motsvarande heltid med tillträde 1 november 2026 eller enligt överenskommelse.   Projektbeskrivning och arbetsuppgifter Malaria orsakas av Plasmodium-parasiter som invaderar och förökar sig inuti röda blodkroppar. Sjukdomen uppstår till följd av att de röda blodkropparna förstörs och att infekterade celler fäster i mikrovaskulaturen inne i kroppens organ, en process som kallas organsekvestrering. För att kunna etablera en replikativ nisch, ta upp näring och utsöndra avfallsprodukter, samt fästa i mikrovaskulaturen, så exporterar parasiten hundratals proteiner in i den infekterade värdcellen. Genetiska screens har kartlagt vilka av dessa exporterade parasitproteiner som är viktiga för infektion under blodstadiet. För majoriteten av de exporterade proteinerna är det dock fortfarande okänt vilka cellulära processer de faktiskt deltar i. Detta projekt kommer att utnyttja Plasmodium berghei, en malariamodellparasit som infekterar gnagare och vars genom kan manipuleras effektivt, för att systematiskt undersöka rollen hos exporterade parasitproteiner vid malariainfektion och sjukdom. Du kommer att utveckla och tillämpa skalbara genetiska verktyg samt nya analysmetoder för att identifiera de parasitgener som kodar för exporterade proteiner av betydelse för parasitens invasion av värdceller, förökning och organsekvestrering. De mest intressanta kandidatgenerna kommer att studeras vidare genom fördjupade studier i såväl P. berghei som människomalariaparasiten Plasmodium falciparum.  Du kommer vidare att utföra experimentellt och bioinformatiskt arbete, designa experiment, presentera dina resultat, skriva manuskript, hjälpa till med handledning av studenter samt aktivt medverka till en kollegial och inkluderande labbkultur.   Behörighet  Behörig att anställas som postdoktor är den som avlagt doktorsexamen, eller utländsk examen som bedöms motsvara doktorsexamen, inom biologisk vetenskap. Salivary gland tumors are a heterogeneous group of tumors originating in the salivary glands of the head and neck region. The prognosis for patients with malignant salivary gland tumors is typically very poor, as there are no effective treatments for recurrent or metastatic disease. The treatments currently used (surgery and radiation therapy) are also often disfiguring from both a functional and aesthetic perspective. The research group has extensive experience studying salivary gland tumors and has identified several driving genomic alterations in these tumors. Duties  We are looking for a researcher to help develop a new targeted treatment for salivary gland tumors. As part of the project, the researcher will plan, design, and conduct research that includes, among other things, the culture of primary/short-term-cultured tumor cells in 2D and 3D models, drug treatment/screening, proliferation/apoptosis/spheroid assays, quantitative PCR, RNA-seq, and immunoblot analysis. The duties also include project management, supervision, and written and oral reporting of research data. Qualifications  A Ph.D. in a relevant field (e.g., biomedicine or molecular biology) is required. Postdoctoral research experience in the field is preferred. Previous experience with cell-based assays, in vitro drug treatment, and the culture of salivary gland tumor cells is highly preferred. The position requires a high degree of precision, independence, and initiative. Applicants should also possess strong collaborative skills and a problem-solving mindset. Om rollen Som Analytical Specialist kommer du att arbeta med utveckling, optimering och implementering av molekylärbiologiska analysmetoder med fokus på PCR-teknologi.
